扩展显微镜(Expansion Microscopy, ExM)由Ed Boyden团队于2015年首次推出,通过将生物样品包埋到具有超强吸水膨胀能力的聚合物中,将样品均匀撑大,使得普通荧光显微镜也能实现超分辨成像。自发明以来,ExM已经用许多不同类型的生物样品进行了测试。然而使用ExM面临的主要挑战是聚合和消化过程中的荧光信号损失。最近开发的标签保留扩展显微镜(Label Retention Expansion Microscopy, LR-ExM)解决了三功能锚(trifunctional anchors)的问题。
